Introduction Hydroxyethylpiperazine ethane sulfonic acid HEPES is a zwitterionic buffer, which belongs to goods's buffer, which has good buffering capacity in the range of pH 6.8~8.2, and can control constant pH for a long time. The concentration is 10~50mmol/L, and the general culture medium contains 20mmol/L4-hydroxyethylpiperazine ethanesulfonic acid, which can have good buffering capacity and no toxic effect on cells. Hydroxyethylpiperazine ethanesulfonic acid has just been widely used in cell culture media, IVD kits and other fields, and is often used as a cosmetic additive because it can control the constant pH range for a long time and is non-toxic to cells. UsesHydroxyethylpiperazine ethanesulfonic acid is often used as a biological buffer, and is often used as a reaction buffer, prehybridization buffer, and hybridization buffer for the separation and analysis of RNA nuclear components; and 3′-terminal labeling of RNA and T4RNA ligases; Or used in biochemical diagnostic kits, DNA/RNA isolation kits and PCR diagnostic kits. It is a hydrogen ion buffer that controls a constant pH range for a long period of time. Application area (1) When used as a buffer: It is used to balance the acidic substances in the cosmetic solution and stabilize the pH range of the product for a long time. (2) When used as an active agent: it can better maintain the activity of microbial fermentation product extracts, so that the anti-aging effect of the product lasts for a long time. (3) When used as a peeling agent: softens keratin and promotes cell metabolism. (4) When used as a permeability promoter: promote the transdermal absorption of various functional components in cosmetics to achieve the effect of brightening skin tone in time. Preparation process (1) Preparation of sodium isethionate: a certain amount of liquid alkali and purified water are put into the addition reaction kettle, the temperature is controlled at 80 °C, and then SO2 is introduced into the reaction kettle to control pH=5.5, and then ethylene oxide is introduced until pH=6.5, and so on until the reactants are completely transformed. After decolorization and filtration, the sodium isethionate synthesis solution was decolorized and filtered, and a certain amount of water was distilled by vacuum steaming and chemicalbook, as well as cooling crystallization and centrifugation, and the sodium isethionate product was extracted. (2) Condensation addition of sodium 4-hydroxyethylpiperazine ethionate: the sodium hydroxyethyl sulfonate:hydroxyethylpiperazine obtained in step (1) is fed and mixed evenly according to the molar ratio = 1:0.8, and the catalyst is fed and mixed according to the ratio of sodium hydroxyethyl sulfonate mass of 2.5%. Through the pipeline continuous production mode, the temperature is controlled at 180°C, the pressure is 2MPa, and the reaction time is 30min. (3) Resin exchange of sodium 4-hydroxyethylpiperazine ethanesulfonate: H732 hydrogen-type ion exchange resin is placed in the exchange column, the reaction solution obtained in step (2) is diluted with water, and passed into the exchange column for ion exchange, so that the liquid level is about 10mm higher than the resin, until the effluent pH=6, and the ion exchange of 4-hydroxyethylpiperazine ethanesulfonic acid is carried out through the hydrogen-type ion exchange resin. (4) resin regeneration: in step (3), the exchanged resin is converted from hydrogen type to sodium type resin, the exchanged sodium type resin is placed in the exchange column, pure water is added to make the liquid level higher than the resin about 10mm, SO2 gas is introduced into the exchange column according to the flow rate of 800mL/min to regenerate the resin, the regeneration time is 8h, the regenerated liquid after the reaction is added to the reaction kettle of step (1) as raw material, and the regenerated resin can continue to be used as the resin in step (3) for exchange reaction. (5) Purification of 4-hydroxyethylpiperazine ethanesulfonic acid: the synthesis solution after the exchange of hydrogen-type cationic resin is decolorized and concentrated by vacuum distillation, and ethanol is added, and then cooled and crystallized for 3 hours, and the crude product of 4-hydroxyethylpiperazine ethanesulfonic acid is obtained after centrifugation, and the centrifugal solution is distilled, and the ethanol can continue to be used for the purification of 4-hydroxyethylpiperazine ethanesulfonic acid, and the distilled mother liquor is passed into the pipeline of step (2) as part of the raw material. 4-Hydroxyethylpiperazine ethanesulfonic acid crude product is refined to obtain the high-quality hydroxyethylpiperazine ethanesulfonic acid of the present invention.
